Principle
Microscopic examination of skin scrapings allows direct detection of pathogenic organisms like fungi and parasites causing skin infections through observation of morphological structures unique to each pathogen.
Solutions & Equipments
Sterile blade/scalpel
Glass slides
Saline/KOH wet mount
Staining solutions (Grams/Giemsa)
Light microscope
Sample Type
Skin scrapings obtained from the active border of dermatitic, vesicular, pustular or scaly skin lesions.
Procedure
1. Clean the sampling skin area with 70% alcohol
2. Use a sterile scalpel or blade to scrape the active border of the lesion
3. Transfer obtained material quickly onto a glass slide
4. Prepare a wet mount with saline/KOH and coverslip
5. Stain additional sample with Gram or Giemsa stain
6. Examine unstained and stained slides under microscope
Results
Microscopic findings – presence of fungal hyphae/spores, parasites, inflammatory cells etc. aids in diagnosis.
Interpretation
Identify fungi (branching hyphae) or parasites (eggs, larvae)
Assess inflammation by noting neutrophil and eosinophil density
Correlate to clinical condition to guide treatment
Notes:
