Creatinine is a waste product that comes from the breakdown of creatine in the muscles and is normally filtered out by the kidneys. in order to measure the creatnine level we need to use some clinical chemistry techniques to do that .

Principle

The serum creatinine test relies on Jaffe’s reaction, where creatinine reacts with picric acid in alkaline medium and forms an orange color. The amount of creatinine in the blood is determined by the color intensity using a spectrophotometer. High levels of creatinine may suggest kidney damage or impairment.

Equipments

1. Tube
2. 1000, + 100 Ul Automatic Pipette
3. Spectrophotometer / Colorimeter
4. Creatnine Bi Reagent

Sample Type

Serum r Heparinized Plasma is Acceptable to perform the Test

Procedure

1. Prepare working Reagent by Mixing equal volume from R1+R2
2. in clean dry tubes add 1,000  μL
3. Prepare the Chemistry analyzer * You will need stop watch if you are using colorimeter
4. using Pipette take 100  μL from serum sample and add it to the reagent *Note that there is no incubation period outside
5. once you added the sample record reading 1 after 30 Seconds and record reading 2 after anothe 60 seconds
6. Now comes the calcuations step

Calculations

Serum Creatnine = Reading 2 of test – Reading 1 of test / Reading 2 of std – Reading 1 of STD x STD Conc

Unit is : mg/dl
Normal Range: Adults : 0.6 – 1.1 mg/dl
Children: 0.4 – 1.0 mg/dl
