Principle
The total WBC count can be inaccurate if there are nucleated RBCs (NRBCs) or other immature cell types present in the blood. A correction factor must be applied to get an accurate TWBC.
Equipments
1. Microscope
2. Oil
3. Thin Blood Film Stained with (Wright Stain like Giems or Lieshman Stain)
4. Counter
5. Pen & Paper
Sample Type
Whole Blood (EDTA)
Procedure
1. Put oil in the thin blood film and use x 100 oil immersion for counting (you can use x40 for correction but you need to put coverslip over the oil to protect the lens from oil.
2. In the ideal area of slide Count 100 white blood cells including the Nucleated RBCs
3. After finishing 100 cells calculate the number of WBCs and the Number of NRBCs
4. Use the following calculation formula to obtain the Corrected TWBCs Count
