Urine Analysis

Principle of the Test

in Urine analysis Test we prepare wet mount sample in slide After centerfugation of the urine which Sediments all the urine components in the bottom of the tube.

Equipments

in order to perform this Test we Need:
1. Fresh Urine sample
2. Microscope
3. Slide
4. Coverglass
5. Centerfuge
6. Urine Strips

Sample Collection Instructions

  1. Urine Sample must be collected in clean and dry Urine container
  2. Urine sample must be delivered to the lab immediately or 1 hour at maximum from the time of collection
  3. give instruction to the patient on the amount of the sample required

Procedure

  1. Fill third of the centerfuge Tube with urine sample and add the balance in the opposite site of the centerfuge
  2. centerfugate the sample for 5 mins @ 1,500 RPM
  3. Dispense the Supernatant back into the Urine Container and Put the sediment in the Slide
  4. Put the cover Slide above the urine sediment in the slide
  5. Using X10 & X40 Microscope Lens Diagnose the Sample *(X10 for observation and general view of the field While X 40 Lens is used for the Dignosis and reporting)

How to write the report

In Urine analysis there are two stages of examination
1. Macroscopic Examination: And this include
A- Physiscal Examination (Color, Turbidity, etc,……)
B- Chemical Examination (Ph, Protein, Sugar,. etc……)
2. Microscopic Examination: Where you use the Microscope to observe cells, parasites, Bacteria, etc….

1. Macroscopic/Physical examination

in this Step we must report:
A- Physiscal Characteristics of Urine Sample and it includes:
1. Color (Normal color is Yellow)
2. Appearance (Normal Appearance is Clear)

B- Chemical Examination

  1. ph
  2. Protein
  3. Sugar
  4. Ketone (Acetone)
  5. Specific Gravity
  6. bilirubin
  7. Blood
  8. WBCs
  9. Urobilinogin
  10. Nitrite

Microscopic Examination

in this Step we Must Report
1. RBCs: Report by Number/hpf
2. Pus Cells: Report by number /hpf
3. Epithelis Cells: (Report By +)
4. Crystals (Report By+)
5. Casts (Report By +)
6. Parasites (Ova, Trophozoite) *Remember to write Parasite Name + Stage + Denssity*
7. Yeast Cells: Report by +
8. Bacteria: Report By +
9. if you have any other Findings you can report it on the others Section
